Description

Summary

PBIO-4D (also known as BPN14770) is a selective PDE4D NAM (Negative Allosteric Modulator) that targets long-form PDE4D enzyme isoforms [-3,-7]. It's selectivity over other PDE4 subtypes and short-form PDE4D is what makes it more tolerable in literature testing than unselective PDE4 inhibitors, not having the same emesis and offtarget risks.

Based on literature [1], it is a potent spatial cognitive enhancer, increasing spatial cognitive measures in healthy Cynomolgus Monkeys and also improving cognitive endpoints in phase trials.

The compound has been well tolerated in its phase trials, with a low incidence of side effects being reported. No serious adverse events have been reported and it has been successful in multiple phase trials for Fragile X.

It also has a favorable pharmacological profile, being relatively potent and with a favorable half life of approximately 10-12 hours.

It is soluble in PEG-400 and DSMO up to 80mg/mL, with poor solubility in water. It can be stored up to 2 years with proper storage.
