Description

Summary

TAK-653 (also known as 9E3TOE5RIZ) is a selective AMPA PAM (Positive Allosteric Modulator) that targets the same pathway as some notable racetams but with higher selectivity and without agonist affinity. The compound has been developed by Takeda (Millennium Pharmaceuticals, Inc.) for treatment-resistant depression, however comparitively, its effect on spatial cognition is more significant and interesting.

Studies have shown that TAK-653 can cross the blood-brain barrier and increase corticospinal excitability in both rats and humans. In addition, it has been shown to improve cognitive functions in multiple domains in rats and working memory in monkeys.

In one study on humans, TAK-653 improved the participants performance on the stroop test, which is indicative of spatial cognitive function.

In terms of side effects, TAK-653 has been well tolerated in studies, with a low incidence of side effects being reported. There were no serious adverse events reported and no participants withdrew from the studies due to adverse events. It also has a good pharmacological profile, being very potent at low doses and with a favorable half life of approximately 10 hours.
